Reliable Determination of Dexibuprofen Content with Polarimetry

Reliable Determination of Dexibuprofen Content with Polarimetry

Rapid measurement of optical rotation for monitoring chiral enantiomeric purity in pharmaceutical quality control

Ibuprofen is among the most widely used active pharmaceutical ingredients worldwide for the treatment of pain, inflammation, and fever. It is successfully applied in both acute infections and chronic diseases. What is less commonly known is that the pharmacological activity of ibuprofen originates exclusively from dexibuprofen, the so-called “active enantiomer.” Conventional ibuprofen is marketed as a racemate, i.e., a 1:1 mixture of dexibuprofen (S-enantiomer) and levibuprofen (R-enantiomer), the latter being largely pharmacologically inactive. This chiral property has direct implications for pharmaceutical efficacy, tolerability, and ultimately the economic efficiency of a medicinal product. The targeted production and analysis of pure dexibuprofen enable lower dosages, a faster onset of action, and potentially fewer side effects. In pharmaceutical API manufacturing, formulation development, and quality control, the precise differentiation of enantiomers is essential. One of the key challenges for pharmaceutical manufacturers, QC laboratories, and contract testing laboratories is the reliable verification of the enantiomeric purity of ibuprofen and dexibuprofen following synthesis. The solution lies in optical polarimetry, a well-established, rapid, and non-destructive analytical technique for determining optical rotation and assessing the purity of chiral substances.

SCHMIDT + HAENSCH Solution for the Purity Analysis of Chiral Molecules Using a Polarimeter

The VariPol laboratory polarimeter from SCHMIDT + HAENSCH enables precise and reliable analysis of enantiomerically pure substances such as dexibuprofen. It is ideally suited for the polarimetric analysis of ibuprofen and dexibuprofen in pharmaceutical quality control, as well as for determining the chiral purity of pharmaceutical active ingredients in regulated laboratory environments compliant with cGMP standards. Measurements are performed at a wavelength of 589 nm, delivering highly accurate values for specific optical rotation. This allows not only the determination of the concentration of the active enantiomer, but also the early detection of deviations in production or formulation processes. The VariPol system was specifically developed to meet the stringent requirements of the pharmaceutical industry. Its modular Aquisys3 software platform allows flexible adaptation to individual workflow requirements and is fully compliant with 21 CFR Part 11. This ensures secure and reliable operation in regulated pharmaceutical manufacturing and quality control environments. Integrated temperature control guarantees stable and reproducible measurement results, even for temperature-sensitive samples. In combination with quartz control plates, the instrument ensures long-term stability and precision of measurements. Furthermore, the broad range of available polarimeter tubes for the VariPol system provides a suitable solution for virtually every application, from micro tubes for minimal sample volumes to convenient flow-through tubes with hose connections for particularly simple and efficient sample handling. Rely on the proven analytical technology of SCHMIDT + HAENSCH for reliable polarimetric analysis and differentiation of ibuprofen and dexibuprofen.
